Frederica,
I used to own the 100-400 and used it with the Canon 1.4x extender. You do lose AF at the long end because the max aperture becomes f/8. Results were OK. I did find the manual focus very hard to get right when shooting birds on the wing, but I guess with a lot of practice...
I haven't tried it myself, but all the reports I've seen suggest that image quality is badly affected with the 2x TC. My experience with the 1.4x is that it does not noticeable affect image quality.

I have the 100-400L and the 2x extender. I also have Both 10D and D60
1. I lose auto-focus at all ranges.
This is the most annoying part as I am very poor at manual focus due to my eye adjusting for the optics (too many years staring into badly calibrated zoom 500 microscopes) and the 10D does not have a good focus screen. My only saving grace is depth of field.
2. I get noticeable degredation of quality, except in very bright daylight, however, I get slightly more detail at the far end 600-800mm equivilant, then I would by cropping and enlarging otherwise.
So based on my experience and from what I have read, the 1.4x is a better buy then the 2x.
