Before I left the UK to go traveling Australia I looked at all the options for a long lens. I nearly got the 100-400 but finally went for the 300mm f/4L IS, a decision I haven't regretted once. I cannot sing the praises of this lens enough. I thought it would be the 24-105 that would become my main walkabout lens but that honour has gone to the 300, it's hardly been of my 20D. Of course it all depends on your main subject interests, over the last few months I've found myself taking mostly wildlife images (birds mostly) but when walking through Australia's wonderful national parks this lens covers all areas, as you can see from the examples below. It's light enough not to put a strain on your shoulder after a long hike and the built in hood is just fab. Of course having the IS is a must if like me you tend to be a bit lazy when it comes to getting the tripod out :)
