What did the upload report?
Check that settings in path editor are correct. Especially that ftp path and URL point to same location on your server disk.
You can do rethumbnail after you change settings.

I assume you have not used EE before?
EE creates all folders needed on upload.
Setup routine should be done in this order:
FTP servers
Server tools
Resize methods
Paths
When they all show green and are set up, then you can try upload - there is no point even trying if some of above are not set up properly. I'll try to explain them briefly:
FTP
You have got ftp server name, username and password from your provider?
Create an new ftp server and put that info into it. Put the same info in "EE Home FTP Server" and save all.
Use "/" without quotes as ftp test path.
If test goes ok, put / as EE home path, save and click "FIND EE ROOT". Wait for the result, the EE root folder is inverted. Copy that to "ftp path to EE root" box and save. Di the tests to see all is ok.
SERVER TOOLS
Here you set up graphics modules you want to use. You must Imagemagick, GD or Netpbm working in order to have EE resize the photos for you. If you do not know Imagemagick path, emty the path and press enter and EE will try to guess it for you. Usually the paths are given in server info email you get when you buy a server.
Test each and click "is installed" if it works, this way you tell EE that this tool is available. GD2 should be always available with newer PHP's.
RESIZE METHODS
Resize methods let you define resized photo sizes to upload. Make what you like and test that they work.
PATHS
In path editor you put all this together in order to tell EE where each size is stored and what size it is and where it goes with ftp and where it is fetched by the browser.

You can install any ftp server available, this is quite simple (for XP): http://www.pablosoftwaresolutions.com/html/quick__n_easy_ftp_server.html
