what are the best manual settings to use with the 400D when shooting people and lights in niteclubs? esp with low light..

There is no such thing. Generally, you want to keep your ISO levels high about 800 or so and a fast lens at or near wide open aperture to keep shutter speeds up. But as lighting condition varies as I'm sure it will, you'll need to change for the dof you want and the ambient light available.
In all honesty, low light shooting in very tough and takes practice to get it right. Take a lot of pictures and use your histogram to adjust your settings.

I would add to the above advice, always shoot RAW, at least that way you will have a little more margin to work with. It is a really bad environment to have to shot in though and I think only practice and keeping a very close on the histograph
