Just received the lens and read the manual but still confused about something. How do you use a macro lens, I mean how do you get the magnification you want. Do you get closer to the subject... the closer the more magnification?
Appreciate any help.

Just get close to the subject. The closest you can get with the Canon EF 100mm f/2.8 Macro is when your subject is about 15cm (6 inches) from the front element of the lens. When you're that close, the magnification will be 1:1.
Note that it's not easy to make handheld photos at such a high magnification. Every little vibration will be visible, and the depth of field is very small. You'll need to stop down the lens to get a larger DOF, but ofcourse then you'll also have to use a slower shutter speed to get the exposure right.
If you want to make photos at 1:1, you really need a tripod, and use mirror lock-up if your camera has it.
