I have a40d, basic two light set up with white umbrella, black umbrella, in sync flasy system, backdrop stand, and some backdrops, tripod with ball head, and two beautiful children. I want to take an awesome pic of them to show off to my family and friends. I have no idea how to go about setting it up, any advice?

I was thinking white light to my left, black reflective stand to the right of kids.

I guess I will just have to play around.

I have no clue what to set my camera on while using manual, any advice will be appreciated then I will post my pic.

Judging by your other posts you are new to photography.
Like many other disciplines it needs a good understanding of the basics and plenty of practice.

http://photography-on-the.net/forum/showthread.php?t=138912 has lots of links to lighting related stuff.

There are lots of dependant variables in your question. Its much like asking:

I have a mixing bowl, a couple of spoons and a gas stove and want to make an awesome cake for my family. I have no idea how to do it, can you help?
