If you're going to be using ratios, then setting the camera to manual would probably work out better for you in controlling the ambient lighting or keeping the shutter speed from going too low if ambient lighting is very low. Of course, you can also control ambient using EC in Av, Tv and P modes but only up to +/- 2 stops (+/- 3 with 1D series).

E-TTL (II) is a way to control flash exposure automatically, as opposed to setting the flash to manual power selection.
On the camera, the various automatic modes (P, Tv etc.) are ways to control exposure to ambient light automatically, as opposed to the M(anual) mode on the camera.
As these things control two different light sources, they are more or less independent of each other, and can be mixed in any combination, as you like.

When using any flash it's better to use Manual Mode because image exposure is easier and more accurately to control..
I have a ST-E2 which I love.. I sometimes use two 430EX Speedlites using the ratio function but the camera is always on Manual Mode and Manual Focus..

St-e2 really rocks.
I only have it for a week but I just love it. It works flawlessly.
I find easier to work in Manual mode and just tell the 580ex to calculate the light I'm asking for with my adjustments.

The only problem that i have with wireless ETTL in my "home studio" is that the exposure changes everytime a little bit. On location ETTL is superb because it's fast and easy.
But at controlled space i don't use it anymore. I like to set up white balance manually, and both flashes manually, and trust that the images will look the same shot after shot.

ST-E2 + Radio Poppers in AV(w/-EC) or M ROCKS!
Really opens up your options for quick, creative, outdoor off-camera lighting. As we know, the ST-E2 alone (without RP's or, perhaps, the new PW's) can really stuggle outdoors.

thank you guys! so as i understand all camera settings will work with sT-E2. in AV/P/Shutter Priority the only way to adjust the flash exposure is using flash compensation and in Manual mode i can do more adjusting Shutter for ambiant and Fstop for flash exposure.
Please let me know if I misunderstood something.

That pretty well sums it up Jun..But beware not to allow the shutterspeed to drop too low..
Do a search on "dragging the shutter"..
