Hi
Will the Canon EF 50mm f/1.8 Mark I lens work on my Rebel 350 XT?

Yes. Any EF lens works on any EOS body.

The MarkI or the MarkII

The only real difference that people care about is the metal mount. Reviews say the MkII is slightly better but they're basically the same. I say save your money and get the MkII for $70 or so. I have it and it's great, but I'm ready for the USM on the 1.4 so I have one on order to arrive this week.
If you're using a lens so much that the metal mount is required, you should be using the 1.4 :)

I went for the used MKI and have to say that I love the lens.
It provides great images and has very decent build quality.
Apart from the rather slow and noisy focus (same on MKII), there's really nothing I don't like about the lens.
I once read a comparison on the web that explicitly compared the MKI version against the MKII version, but could not find it.
Here are some of the links I found, nevertheless:

If you can get a Mk.I version for a decent price, it's a great buy.
However, with the crazy prices I've seen on eBay, typically >$150 for mint, save your money and just get the Mk.II version or better yet, go for the 50/1.4.

Go for MKI if you can (I agree w Andythaler & thomasrhee). A little noisy, but what a lens! One of my absolute favorites. No wonder it currently holds 1st place with 31% of the votes in the polll Vote for your Favorite -=Starter Lens=-, and also no 1 in the poll Top 10 Portrait Lenses (in this section of the forum).

try to get the 50/1.8 attached to an EOS 650 body. Those usually sell for a 100 or so, and you get the camera as a bonus.
Cheers
