I am considering purchasing ST-E2 to trigger my 580EX2 and 550EX. If my 580EX2 is inside of an Apollo softbox, will the ST-E2's signal reach the flash? Thanks.

Not sure about that, but as long as the IR sensor can get signal you should be fine. Try it out and let us know. :D

If the flash is inside the softbox, the only place it'll be able to receive any command signal from the master is via the front diffuser panel so you'll have to make sure that the master is pointed directly at the front diffuser panel (and the slave IR sensor would need to be pointed towards the diffusion panel too). If the body of the flash is outside the flash, then you'll need to position the body so that the IR sensor is pointed towards the master (for direct line-of-sight) or at a wall or ceiling where the signal from the master can be bounced off of.

You have to rotate the head so the front of the body faces your camera.
