Lighting and DOF look great. Couple things to take into consideration. wife's left hand is cut off while her right hand is growing out of your nieces' stomach. Also cut her feet off. Not sure what is causing her right lens on her glasses to look cloudy must be some type of glare. All in all i think this is a wonderful family portrait shot that would be worthy of framing. Composition is great.

Exposure / lighting is great.
I THINK their faces are very slightly out of focus or perhaps just need some sharpening in PP. I think for shooting two people like this F4 is a little bit shallow for DOF.
The right lens of her glasses seems to have caught some glare. The frames of her glasses intersect the eyes.
Childs feet are cropped. Wifes arm is cropped at the wrist.
You have a cool backdrop and I can see some moving water in the bottom left of the frame. Either include more of the moving water in the composition, or exclude it entirely.
I still think the photo is good and most non photographers would be pleased with it :)

Thanks what would be a good F/stop to use for a situation like this?
thanks

that depends on your focal length and your distance away from the camera. 5.0-5.6 maybe. 4.0 is too shallow, your wife is completely in focus your niece isn't it.
The shot is beautifully exposed. I'd crop it a bit different. The hand out of the stomach is not to bad. but your wife's other arm seems awkward at the cut off.
I think the feet would have also added something.. you have room to crop on top...make them the primary standout.
Good attempt

Here is a different crop that I tried, the feet don't bother me so much. I like the two people are really the main aspect. The hand still bothers me a bit but not as much.
