I have a Canoscan FS4000US film scanner and I think I need to tweak the focus just a bit. Does anybody know how to do that?

In the software interface, there is a control for the focus. Did you try that? I tried some different manual settings and came to the conclusion that it does it automatically better than I can pick it.
I suggest you try a manual setting one way or the other from whatever the automatic setting was suggesting. It is possible for film or a slide to be "sloping" and not perpendicular to the sensor aim, but I think that is rare, at least with 35mm slides, which is what I use on it. I've seen film that was curved, and obviously that is going to play hell with the autofocus.
