Send your model to the gym for a programmed workout.

Have the model tan. The darker the person is, the larger and more defined their muscles look. Also, have them oil themselves up. All professional bodybuilders tan and oil up before a comp to help give contrast to their muscles. Shaving is another big thing. Hair hides the muscle beneath it, so if the model is clean-shaven, they will look bigger. Also, fool around with the lighting. Thats another big thing because shadows can also make the person look more cut and defined depending on how they fall on the body. Last but not least, have them pump up before the shoot. That is as simple as doing a mini workout right beforehand to get the blood pumping into the muscle tissue. It can be as simple as doing some bodyweight exercises, to 15-20 reps of an exercise for each muscle group.

In addition to the above 1-2 days with minimal fluid intake increases the definition.

If you have CS4 you could use the liquify tool.
