I've not tested the two, but luminous landscapes have

Sharpness wise, the 400 F5.6L gets the point. Aparrently the prime also manages to AF quicker too. As for IS, get your crystal ball out ;)

I own both. The 400 f/5.6 autofocuses much faster in low light, low contrast situations. The 100-400 gives you much more flexibility and very sharp pictures. My 400 has gone back to Canon 3 times for soft focusing and a dirty interior lens element. I don't think it's an eithor or situation. I use them both in very different situations.

I had the 100-400mm, it was returned, and was swapped for a Bigma, as it had "issues". It had a 50 foot backfocus, and was to soft to even properly post process. Then there was the push/pull zoom and zoom lock, which was incredibly annoying. I guess you could say that I did not like the lens. Maybe one day I will try another.

Now the 400mm 5.6 is an awesome lens. I wish it had IS too, but I can live without it. It is incredibly sharp corner to corner, and is really not that bad in the weight department. It is lighter then my 300mm f4L IS and 70-200mm 2.8L IS. Great birding/wildlife lens with the 1.4x TC.
