It should work, try it with lots of light and nearby opjects. How is your camera set to lens drive when AF impossible? If off then try on, it may be to much out of focus to start with.

Thanks! Your 100L and 25mm extension tube should work together... just make sure that you are working within a few cm of the minimum focussing distance, because it won't focus beyond that.
