Oh yes, tripod mounted definitely.
I had the Canon (EOS R) + (RF-EF Adapter) + (Helicon FB Tube) + (Ext Tube 25mm) + (EF Extender x1.4 Mk III) + (180mm F3.5 L) so quite a long imaging train. I find the AF on the 180mm slow, but once you have it dialled in on the subject, it is accurate and responsive. If a gust of wind blows the subject out of the frame, the AF winds all the way out to infinity and I have to manually wind it back to prevent it hunting.
This was a stack of some 30 frames so IMO, a tripod is essential for this setup.
The EOS R articulating screen is a joy to use; I just "Touch" the closest point of focus (on the berry in this case) and then press and hold the shutter and off it goes.
The 180mm + Extender x1.4 gives the 252mm focal length and the 25mm Tube+ Helicon FB Tube allows me to get a little closer.
