At a glance seems a little soft (maybe even missed focus a little bit? could also be jpg compression, though...) and seems like the background needs to be more separated from the subject. So either a wider aperture or a bigger distance between the subject and the bg.

I can't say much in detail about the lighting but maybe someone else here can.

This is a good start, but if I may offer a few suggestions... First, your lighting appears to be too even and it is a little underexposed. Leave one light at the same power and boost the power on the other (key) light. I'd also raise the key up to get the catch light around 10:00 or 11:00. Also, try turning your wife/model so she isn't so square to the camera. Try different angles from both the left and right. Lastly, try shooting from a slightly higher point of view. Not too high, but have the lens at about eye level or slightly higher.

give it another go and post the results.

The head on shots are usually never flattering, have her turn 45* to the camera and look back at you next time.

Too much blue in the photo.. The shirt blends in too much with the background screen and in my opinion maybe a different color background would have been better. I don't have enough experience to comment on much else since I am still learning about photography and I like to do mostly nature/wildlife photos, but that is just what stood out in my mind when I first saw the photo at a quick glance.

Several points: some have already been mentioned.

First have her turn slightly left or right. This will make her look thinner, and I've never known a woman who didn't want to look thinner, no matter their size. Also, with less even light, (think Rembrandt) some shadowing will also help this.

And as mentioned, shoot from a higher angle. More flattering to your subject. Thankfully, you have a willing subject. And lastly, perhaps you could have her more engaged? Maybe a laugh or something that will look less like a mug shot? Nice looking lady that could look nicer with a few small changes. ;)
