One question. Why would you want ALL of that in one backpack for your back? That would be awful for it! I have a >> Case Logic (external link) <<. When I fully load it up(Gripped 50D, 70-200mm 4.0, Sigma 17-70mm 2.8, Sigma 10-20mm 4.0, 430 ex2, Vagabond lithium mini, other little bits), it feels heavy when you pick it up until you put it on your back. Once on your back its very well supported and has a much lighter feeling contrary to the POS canon backpack thats was a waste of money(straps were 10% away from ripping out of the top of the back.) Holds up to a 16" or 17" MBP.

Best gift my gf bought me and would recommend to anyone.

Well if I am going on a trip, I want to be able to choose between all of my gear. Usually I go for 2-3 weeks during the summer holidays and do a wide range of stuff. One day I might use the 400mm for wildlife and the other day the 10-20 for architecture or the 70-200 for a theatre show or for a sports contest. If you are traveling by plane it is better to pack everything in one bag that you can take on the plane, this way your gear won't get damaged.
And I don't think it is bad for your back as long as you use a good backpack with a strap around your hips. I have walked with a good backpack (Haglofs bag) with my tent, food, clothes... in. Anyway, I won't be using the big backback with everything in it when I arrive at my hotel. I will choose everyday which gear will I need today and then put it in a smaller bag (my current Flipside 200).

I find the Gura Gear to be much lighter and I can stow away all the straps so it doesn't dangle around when I put them away in the plane. The bag can also carry an insane amount of gears, everything you listed and more. Gura Gear thought of everything when they design this bag. It is a little expensive but I think it's well worth it.

I've been searching for a carry-all bag also... My front-runner is the Kata MultiPro-120 PL... but it is so new that I haven't seen any reviews on it. But who knows, I may swing back to a "safer" choice and go with a ThinkTank.

I have a vertex 300 aw and love it. It is a big old beast, but very comfortable even when fully loaded.

reading this, my concern was about flying and leaving any camera gear in the room. What do people do to secure it? Ensure they have a safe or check it at the front desk?

I wouldn't trust placing it in a dresser or unattended luggage...

I either lock it in the safe or if safe is too small, I'll just lock in in my suitcase. If you are really concern then lock in in the suitcase and check it in the front desk.

With all the equipment you have, it WILL be expensive. Otherwise you cheap out(like the canon bag I had) and it falls apart on you very quick and you're left stranded having to spend even more money. That bag I showed you should fit most of everything you have + a laptop up to 17"
