Any Mechanic In The House?

My go to big ride for Photography job when I need to haul most my gears Toyota 4Runner V6 2WD reached 100K. Please advice what are things that I need to do get a total maintenance done.
I just want to estimate the total cost also see what are some simple things that I can DIY.

100k these days is more of a psychological milestone than anything. I don't have a lot of direct experience with Toyotas (Volvo/VW Mastertech) but from what I've seen I wouldn't be too worried. Have you been following the recommended maintenance schedule for the vehicle? Beyond what's recommended in the "severe driving" schedule I say find a competent, trustworthy tech and listen to what he tells you. Avoid cheap "quick lube" stops. Make sure the guy that handles your vehicle is your go to person for everything. Oil changes give a tech the opportunity to scrutinize a vehicle, looking for anything that may be wearing out, broken, leaking, etc. It really is all about being punctual and thorough with your maintenance, and using someone you can trust; preferably a specialist, preferably not the dealer.

@100k Recommend Normal Service is Oil/Filter Change and Flush Cooling System with a bunch of checks. -Easy to do with minimal mechanical ability.
Spark plugs and air filter are due every 30k miles. -Fairly easy.
Valve clearance is checked every 60k miles.

These few things are the big ones on the factory maintenance list. Of course your best course would take it to a competent repair shop and have a vehicle inspection done to see what else may need to be done. Such as brakes, hoses, belts, or any other potential problems.

My qualifications, for what it's worth, ASE Master Certified with L1, L2 & M1-3 Certs. 10 yrs in Audi/Porsche/Mercedes dealership and 12 yrs in independent import shop. Currently I own my own auto repair shop oriented towards imports.

If it's never been done, not a bad idea.
