I am contemplating buying a Nissin Speedlite Di866 Mark II.
My question is - does the auto focus assist lights cover ALL focus points on a 5DII.
This is important to me as I shoot a lot of weddings and many shots are in portrait mode and I always use the top focus point.
I prefer to do this than use the centre point and then re-compose.

Is there nobody out there that can answer this simple question?

Mate,, I doubt there'd be many people out there using this combination, but I have a Nissin Di866, the Mark I.

I just did a quick test using my 50D/EF 85mm f/1.8 and far right focus point, in very dark conditions. No problems focusing.

Should be OK with your 5D and Mark II.
