I am wondering whether anybody has the experience with these 2 backpacks. I need something comfortable for 5D MKIII, 24-105, 70-200 II and possibly another UWA lens. I also carrying a tripod and some other accessories.

Which of these two is more comfortable for longer walks and is more practical?
Thanks.

I have the Vertex 200 AW and can highly recommend it. It has a very good harness system and I find it very comfortable. Tripods secure well unless you are using heavy heads when they can flop about a bit. However if you put the tripod on the back of the pack, rather than the side, there is an extra strap that reduces this problem. Being taller the 300 AW would probably not have this issue.
Do you need such a large pack? I carry a 1D4 + Canon 300 F2.8 L IS, 24-105, 17-40, extenders, filters, spare battery etc easily in my 200 AW which is smaller, cheaper and lighter than the 300. However, if you are looking to expand (500 F4?) your gear then the 300AW is an excellent pack.

Life is for living, cameras are to capture it (one day I will learn how!).

I have the Vertex 300 AW and have been using it for about 6 months. It is a very large bag, but very comfortable to carry.

This pack carries everything in my sig line except the 10d and the 120-300 2.8. It also carries a Hoodman loupe, 1.4 extender, cables and cords, bulb blower, (2) LP-E6 battery chargers, filter system, extra CF cards cleaning supplies, and the pack is not overfull. All together it weighs about 32 pounds full.

I carry an Induro AT313 tripod (28" long),on the rear of the pack when hiking. The shoulder pads have plenty of padded length, unlike my older large Tenba pack, which always cut into me a little. As far as fit, it's very good and I'm 5'11" with a long torso. The waist belt sits a little high on me because of this, but doesn't make it uncomfortable. In fact, most of the time I don't even use the waistbelt on short hikes.

The pack has a weather resistant main zipper, and a rain cover that stows in the bottom, along with a zipping cover to cover the shoulder straps, when carrying the bag instead of wearing it.
