First off, I assume these two are compatible from what I found online. I was wondering if when using the release does it override my current setup using back button focus? I guess I'd still like to be able to use BBF and just use the release to trip the shutter but not sure if this is possible.

If you're set to BBF, the remote release won't cause the camera to focus when you go to trip the shutter - you'll have to BBF then hit the remote, which sort of renders using a remote pointless since you use it to avoid directly touching the camera.

I am looking at using the remote while doing some lens calibration along with my body. I know I could probably just use a shutter delay of 2 or 10 seconds but I thought I might use the remote for that purpose.

Just return AF to the shutter button for your testing purposes using a remote release, that way you don't need to touch the camera for the focus operation. If you want the AFMA value written to the EXIF then you need AF to be active when the shutter is released. BBF gets you nothing if you're doing AFMA on a tripod and it only takes 5 seconds to change the setting.
That's the way I used to do things using DotTune or eyeball test before I bought FoCal.
