An extension tube has greatest effect with a shorter lens, in terms of getting you closest to the subject...a 25mm tube on 50mm is adding a lessened ratio of
[tube extension] : [focal length]
25 : 50 = 1:2
25 : 100 = 1:4
For example, 50mm tube with 50mm lens can get you a 1:1 magnfication image on the film/sensor.
Added to the 25mm, it can give you macro magnification; added to 100m, it can merely get the lens to focus closer than without the tube, but not necessarily macro magnification. For example, I use 25mm tube with my 150mm lens on my medium format camera, to shoot headshots of models and actors/actresses.
