Define "unique"?
RGB = 3, or 256X256X256, or...
What's the point?

I think he's looking for a count when not all 256x256x256 are represented. Just because an image can contain all those possible colors doesn't mean that it does. Or am I missing something?

OK, can someone tell me just what the possible uses are of that measurebating?

Well it certainly gives on an idea of color loss when comparing processing in 8-bit versus 16-bit, for one.

I didn't say it was useful. ;)

Actually, like Scottes said, it could possibly be used to view the effects of any particular editing mechanism. How much color do you lose through repeatedly saving a JPG? What's lost between 16-bit and 8-bit? I've never tried (or even really thought about) any of these, but I'm sure smarter people than me have found a way to put the information to use.

I'm not sure that the number of colors is the right criteria because some combinations may disappear but some new others may appear...

True, but depending on the processing done there will almost always be less colors in 8-bit mode regardless of the number of new colors created. From my tests in the past - with some standard post-processing procedures - the difference is significant.

Here's some "enlightenment" for you, SgWRX: If you convert all your images to GIFs, you can be sure how many colors there are in them! I recommend going for 8, 'cause you can count them with only two hands?

IMAGE: http://photo.klein-jensen.dk/smilies/yield.gif

Here's a simple concept; "Start with more = end up with more." & you can stop sweating the small stuff.
Take 2 identical shots, one in RAW & one at max jpeg. Use a tripod to be sure each shot has the exact same information in it. Convert the RAW file to jpeg. Look at the two file sizes.
A max jpg from my 20D is 2,754 KB. The exact same shot with the jpg extracted from RAW is 4,315 KB which is 1.57X larger.
