this is quite easy in PS...
Add a desaturation layer and desaturate
add a curves layer with an S curve
Add a colour balance layer, or selective colour and resaturate the colours you want

to get it really moody like 300, add a vignette and grain, on seperate layers..

play till happy :-)

I watched the movie "300" last night and the gloomy effect on the entire movie was great. How would I achieve this? I'm sure they shot the entire movie with a filter on the camera, right?

They may have filmed with a filter but that could seriously effect the post production work on trying to key out the Blue/green screen. To me having worked with a compositing program(Apple Shake) the overall look was IMO done in post. If you are looking to recreate that effect.....its not just PP... you have to also achieve similiar lighting when you take the picture. :)
