Is there a difference between modifying the Standard mode (let's say, putting sharpness to 7), and using the User Custom Setting ? (User Def. 1.)

Probably only to save time when you pp, or have a lens which isn't the sharpest.

No, I think you can pick Standard picture style and then increase the sharpness in custom setting as well.
