300x is a UDMA card and you wont see those speeds in your camera. Also, the only way youll get UDMA transfer speeds of 300x on your computer is with a UDMA card reader.

Will the 40d use the UDMA card? or will it not work in the camera?

It will work but you’ll see no benefits, no speed increase.

What we see depends mainly on what we look for.

So the transfer to the computer is faster? A UDMA card reader on USB faster than a standard USB reader?

Whats a good UDMA reader for USB 2.0?

Thanks for the quick responses!

The readers I've seen are Firewire/IEEE 1394, probably Firewire 800 which is faster than USB 2.0.

Well, 133x is enough in my experience. However you use a card reader, 300x will have some extra benefits.
