I just got this lens today. I've only used one other macro lens (the 50 f2.5). What are some of the characteristics of this lens...where is the sweet spot? What is the best way to use this lens?

Sweet spot is probably around 5.6, but this lens is great even wide open. I used it recently for a wedding, and with the USM its great for medium telephoto. 2 x 5Dc, XE-1

You can get to 1:1 magnification with the 100, whereas the 50 only enables 1:2. Very sharp even widen open, but as you get closer to your subject you will need to stop down to increase depth of field. Sebastien
