Well you can switch the operation of the AF-ON button and the * button, so I have them switched around for my purposes for the reasons Peter mentioned but basically it's inconsequential.
So I use * for focus, but what I use the AF-ON button for is the awesome registered shooting mode, and I have that custom function in My Menu so I can quickly register new settings - what it does is it lets you register a bunch of settings, and pressing the AF-ON (or *) automatically switches to those settings, focuses, and lets me shoot. SO for example, if I'm out birding, I register 1/1000s, f/5.6, whatever ISO I need for the given light (M mode), and AI servo, so even if I'm shooting a stationary bird, I can hit AF-ON and go instantly into bird in flight mode if something flies by.
If I'm in studio, I just register default settings (f/11, shutter sync speed, ISO100 etc), so if I'm playing with DOF and the model hits a pose or expression I like, I can quickly switch to something that will give me max DOF and is set up for my lights already.
