I have a 50D and when in manual I cannot change the aperture anymore. I have double and triple checked that it is "all of the way on"/to the second click. Still can't change. I can change in Aperture priority mode. I can use the selection wheel for other things.
I've changed lenses and double checked that I am getting a full reading form the lens to the camera. I have a feeling I messed with a setting today somewhere? I have removed both batteries and left the camera long enough to completely reset, reinstalled the batteries, set time and date-still nothing. Help?

Alright, I just put my 50D in M. I changed the aperture by turning the on switch all the way up to that wierd L setting, it's past the on setting. Then turn the wheel on the back. Did you do that?

Yep, I double and triple checked that it's all of the way to the check mark and then turned the wheel. I shoot all manual all of the time, so I am pretty well versed in what I need to do, but this one baffles me.
I did shoot outdoors today for a fire-from a mega distance so heat isn't the issue. It was snowing lightly, but not enough to get the cam wet. Lenses work fine on my older body, so it's not any of my lenses. I am guessing maybe it's not registering that I am turned on ALL OF THE WAY to the check mark?

Yeah, it sounds to me like it's not registering, like you said. If it worked right earlier and not now, that ain't good.

You have the jog wheel locked. Move the switch back to the on position.

EDIT: Just noticed your second post. Are you using the jog wheel on the back to adjust the aperture in M mode? In Av mode the wheel on the top adjusts, but for M mode it's the wheel on the back. Other than that, the switch may be broken with wheel stuck in the locked position.

It doesn't physically prevent the wheel from moving, but it will prevent it from making adjustments (to prevent accidental adjustments). Line the switch up with the white line that points to the wheel itself, which should allow the wheel to function. If that still doesn't work, then something may be wrong with the switch.

OK, I did a reset of all of the custom functions, now it works. What in those custom functions would lock my aperture? I did change settings around a bit today messing with the d-lighting and some of the picture styles... trying to really push the limits of the different settings, etc on the 50D... I got some awesome shots, but obviously somewhere along the way I locked myself in at f/8. Does anyone know how I managed to do this?
I intend to go to bed with the manual tonight because I know there's something in the custom functions that I changed, but I am sure you guys all know how "informative" the manual ISN'T! Advice?

That's ok, I kinda figured people would assume I was new to the game! I feel like an idiot at this point cuz I have NO CLUE what the heck I did and I THOUGHT I knew my cameras pretty well! Guess I thought wrong!

Glad you figured it out! I'd be interested to know what you Cfn setting you changed as well.

There is a Cfn on some Canons that changes the rear wheel (Quick Control dial
in Canon-speak) function from the default (aperture/exposure compensation) to selecting the AF point or the ISO or several other choices. I don't have a 50D handy, but on the 1D mkIV, it's Cfn IV-3.
Don't be too hard on yourself, with all the Cfns on DSLRs, it's very easy to set something unintentionally. I carry a copy of the Cfns list with me for this very
reason, has saved me a few times.
