Well, at the risk of being lectured by Roger, and to be brief, humans use 100% of their brains in the sense that all structures and functions of the human brain are utilized at any one point in the lifespan of a human being. A person, however, may not use the whole potential of his brain ("Steven, think before you post!"), and at any one time a person does not necessarily utilize all brain structures and functions at once. That's easy to imagine when you think of sleep. Different regions of the brain become active during sleep as opposed to during waking hours. This is the sense by which we don't use 100% of our brains.

I might not have the terminology down right to scientific standards, but that's the gist of it.
